چکیده

Background—Salicylates may have direct vascular effects by mechanisms that are independent of platelet inhibition. Methods and Results—We investigated the effect of salicylates on vascular smooth muscle cell (SMC) proliferation in response to platelet-derived growth factor (PDGF) in vitro. Salicylate concentrations of 5 and 10 mmol/L inhibited serumor PDGF-induced SMC cell count and [H]thymidine incorporation by 62% and 81%, respectively. There was no evidence of cellular toxicity or apoptosis as determined by trypan blue exclusion and FACS analyses. Because cell cycle progression is regulated by hyperphosphorylation of the retinoblastoma (Rb) protein, we examined the effects of salicylate on Rb hyperphosphorylation. Treatment with salicylate, but not indomethacin, inhibited nuclear factor-kB activation and completely abolished Rb hyperphosphorylation in PDGF-treated SMCs. This effect was associated with a decrease in cyclin-dependent kinase (Cdk)-2 and, to a lesser extent, Cdk-6, but not Cdk-4 activity, without changes in Cdk-2, -4, and -6 and cyclin D and E protein levels. Because Cdk-2 activity is regulated by the Cdk inhibitors p21 and p27, we studied the effects of salicylate on p21 and p27 expression. Treatment with salicylate prevented PDGF-induced downregulation of p21 and p27 but not of the Cdk-4/-6 inhibitor p16. Conclusions—These findings indicate that high doses of salicylates inhibit SMC proliferation by cell cycle arrest at the G1-S phase and suggest a beneficial role for high-dose salicylates in the treatment of vascular proliferative disorders. eNOS gene transfer to vascular smooth muscle cells inhibits cell proliferation via upregulation of p27 and p21 and not apoptosis.

OBJECTIVE Smooth muscle cell (SMC) proliferation is a critical component of vascular diseases such as atherosclerosis and restenosis. Nitric oxide (NO) donors and gene transfer of endothelial nitric oxide synthase (eNOS) have been shown to inhibit SMC proliferation. NO may cause this effect by delaying cell cycle progression and/or induction of apoptosis. The aim of the current study was to exa...

TZDs inhibit vascular smooth muscle cell growth independently of the cyclin kinase inhibitors p21 and p27.

The thiazolidenediones (TZDs) are commonly used to treat hyperglycemia in type 2 diabetes. Diabetes is associated with macrovascular disease, leading to accelerated atherosclerosis caused by aberrant vascular smooth muscle (VSM) cell proliferation. Although VSM cell proliferation is inhibited by the TZDs, the mechanism of this effect has not been established. Because of reports that the cyclin ...

Role for p27 in Vascular Smooth Muscle Cell Migration

Background—Rapamycin is a potent inhibitor of smooth muscle cell (SMC) proliferation and migration. Rapamycinmediated inhibition of SMC proliferation is associated with upregulation of the cyclin-dependent kinase inhibitor p27. Previously, we showed that mixed embryonic fibroblasts obtained from p27(2/2) mice were relatively rapamycin-resistant, suggesting that p27 plays an integral role in mod...

Cyclin-Dependent Kinase Inhibitor p27, But Not p21, Is Required for Inhibition of Hypoxia-Induced Pulmonary Hypertension and Remodeling by Heparin in Mice

Heparin has growth inhibitory effects on pulmonary artery smooth muscle cell (PASMC) in vitro and in vivo. However, the mechanism has not been fully defined. In this study, we investigated the role of cyclin-dependent kinase inhibitors, p21 (p21) and p27 (p27), in the inhibitory effect of heparin on PASMC proliferation in vitro and on hypoxia-induced pulmonary hypertension in vivo using p21 and...

BMP-2 inhibits proliferation of human aortic smooth muscle cells via p21Cip1/Waf1.

Bone-morphogenetic proteins (BMP)-2 and -7, multifunctional members of the transforming growth factor (TGF)-beta superfamily with powerful osteoinductive effects, cause cell cycle arrest in a variety of transformed cell lines by activating signaling cascades that involve several cyclin-dependent kinase inhibitors (CDKIs). CDKIs in the cip/kip family, p21(Cip1/Waf1) and p27(Kip1), have been show...
